Analysis & QC Instruments
GenCore is equipped with several auxiliary instruments for assessing sample viability, quality, and concentration.
The Agilent Bioanalyzer 2100 is a micro-fluidics based platform that can analyze the quantity, quality, and characteristics of small RNA, DNA, protein and cell samples.
GenCore has two Agilent Bioanalyzer 2100 systems:

- 8th Floor Brown Building
The Agilent TapeStation 4200 is a high throughput automated electrophoresis tool for analyzing nucleic acid sample quality and fragment length. It offers scalable throughput, is fast and easy to use, and provides researchers with great reproducibility.
GenCore has two Agilent TapeStation 4200 systems on 8th Floor, CGSB.
The Amersham Typhoon RGB is a robust and high throughput biomolecular imager that can be used for fluorescence scanning, phosphorimaging, digitization and chemiluminescence experiments.

The CFX96 qPCR instrument is a powerful and flexible system that combines advanced optical technology with precise temperature control.

The Bio-Rad ChemiDoc is an imager that can visualize and analyze proteins and nucleic acids a wide range of gels and blots, including western blots. The ChemiDoc can analyze stain-free gels and blots, detect chemiluminescence, or perform multiplex fluorescence analyses using a wide variety of gel stains (such as ethidium bromide, SYBR, and Coomassie).

The Life Technologies Countess II FL is an automated cell counter with florescence capability. This allows for cell counting, fluorescent protein expression monitoring, and measuring of cell viability.

The NanoDrop Spectrophorometer allows the quantification of microvolumes. The Department has a number of these units, including one in the Shared Equipment Core, for general use.

The QuantStudio 6 Flex Real-Time PCR system contains a 384 well block and is capable of performing qPCCR, Genotyping, Melt Curve, and Comparative Ct experiments, plus other applications. The system supports SYBR Green and TaqMan reagents, among others.

The Roche LightCycler 480 system consists of a thermal cycler used for rapid real-time quantitative PCR and SNP detection, a computer to control the thermal cycler, and software for the design of probes for hybridization probe analysis and primers for SYBR green analysis.

Flow Cytometry
There are two high throughput flow cytometers available to sort and analyze various cell types using fluorescence. Technician run cell sorting is also available for researchers.
The FACSAria is a high throughput cell sorter capable of sorting 50,000-70,000 cells per second in up to six channels and into four flow streams. The system contains blue, red, yellow, and violet lasers for excitation of a variety of fluorophores utilizing about 10 channels.
**Please note: This instrument will be retiring in FY25 and all experiments will be migrated to FACSDiscover S8.

Quick Reference Guide (with laser configurations)
The Cytek Aurora is a spectral flow cytometer equipped with 4 lasers (V/B/YG/R) and 48 detectors, and is capable of detecting the emission full spectra of samples.
*PLEASE NOTE training for this instrument requires at least 48 hours notice before the training appointment. An experimental form must be filled out and controls and/or test samples should be brought to the training. Contact GenCore for additional information.

The NanoCellect Wolf G2 Cell Sorter is a benchtop cell sorter capable of bulk cell sorting or single cell sorting into 96-well or 384-well plates. This instrument is equipped with 488 nm and 561 nm lasers and can process up to nine colors. This is a self-serve instrument once training is complete.

High-Throughput Instruments
GenCore has a number of instruments used to perform various repetitive tasks in high throughput, including NGS library preparation, normalizations, and cell-based fluorescence assays.
The 10X Genomics Single Cell Chromium Controller is a benchtop instrument for single cell capture and gene expression profiling. The 10X Chromium can capture hundreds to millions of cells with a simple workflow, and users can go from cell sample to sequencing library in less than one workday.

The Agilent Bravo Robotics system is a high throughput liquid dispensing robot. It is equipped with a 96 pipette head and a flexible deck that can accommodate Agilent NGS Option B protocols for library preparation, plate stamps, PCR setups, and much more.

The Formulatrix Mantis is a benchtop liquid handler that offers the lowest dead volume of any liquid handler on the market using its patented microfluidic dispensing technology. It is extremely flexible and easy to use, saving time and money.

The Tecan Spark Microplate Reader is a multimode plate reader that can be used with various plate types ranging from 24-well plates to 384 well plates, allowing scalability for experiments. It is equipped with a high speed monochromator aiding in absorbance, fluorescence, luminescence detection, and more.

The Sage Blue Pippin an target a wide range of molecular sizes (90bp to 50kb) for use in preparing NGS libraries. 5 samples can be run in parallel with no risk of cross-contamination.

Sonicators
There are three different types of sonicators available to researchers for various types of experiments including DNA shearing for NGS, chromatin shearing, tissue homogenization, and more.
The Covaris S-Series Focused Ultra Sonicator is a versatile, high power system that performs at high frequencies for a variety of applications.

The Diagenode Bioruptor Pico sonicators can uniformly process multiple samples and can be used to shear DNA and chromatin as well as homogenize tissue.
